EQUIPMENTS

Major Equipments

Available equipments include 2 laminar-flow biological hoods for cell culture, -80°C and -20°C freezers, 1 high speed centrifuge (Beckman Coulter Avanti J-E high-speed centrifuge) with a variety of rotors, 2 incubation shakers (1 ThermoFischer SHKE8000 series MaxQ orbital shaker, 1 Innova 4230 refrigerated incubator shaker) and numerous small equipment for gel electrophoresis, electroporation systems (2 Eppendorf electroporators 2510, and 1 Bio-Rad Gene Pulser), sonication (Branson sonifier 450), a French Press (ThermoFischer) and many small devices.

For purifying recombinant proteins purifications units are available (1 AKTA Explorer from GE-Healthcare and 1 NGC Chromatography system from BioRad).

Two Methrom 718 Titrino pH-stat are used for lipolytic enzymes characterization.

One ChemiDoc MP Imaging system (Bio-Rad) for analyzing SDS-PAGE, Western Blot, fluorescent assay and a 2D gel electrophoresis system (Protean i12 IEF System) offer large possibilities for many proteomic studies.

One CAMAG Linomat 5 semi-automatic TLC/HPTLC sampler with its Derivatizer (i.e., an automated spraying device for reagent transfer onto TLC/HPTLC plates) and a TLC Spray Cabinet for revelation.

One Tecan SPARK 10M multimode microplate reader, 1 BioTek PowerWave 96-wells microplate spectrophotometer, and 1 ThermoFischer Fluoroskan Ascent 96-wells microplate fluorometer allow the screening of lipolytic activity, lipase inhibitors, mycobacteria growth inhibitors...

The team has recently acquired a Leica TCS SP5 II microscope (Leica Microsystems) equipped with Acousto-Optical Beam Splitter (AOBS), 4 lasers including 405 nm, 488 nm, 561 nm, and 633 nm, and a HC PL APO CS2 63×/1.40 oil objective. With its custom-made environmental control chamber that provides 37°C, 5% CO2 and 20-30% humidity, our system allows to perform 4-colors high-resolution live-cell imaging.

BSL-2 laboratory

A BSL-2 laboratory fully equipped is located in a specific room and managed by an controlled access for studying M. bovis BCG, M. tuberculosis avirulent strain, as well as non-tuberculous mycobacteria (M. abscessus, M. marinum....)

Shared equipments at CNRS campus

Core facilities of IMM platforms center: mass spectrometry (MALDI, ESI-Q-ToF, LTQ-Orbitrap), N-terminal protein sequencing, Electron (FEI Tecnai G2 TWIN 20-200KV) and confocal (laser scanning confocal Olympus microscope FV 1000) microscopes; and transcriptomic analyses.

Two Beckman ultracentrifuges are also available at the Institute.

Lastly, the IMM possesses a specific room to work with radioactive compounds.
